慢性乙肝患者外周血单个核细胞凋亡及细胞因子检测的意义

To investigate the significance of PBMC apoptosis and Th1/Th2 cytokine in the pathogenesis of chronic hepatitis B The apoptosis was measured by propidium iodide staining and the IL-2、IFN-γ、IL-4、IL-10 of PBMC induced by PHA was detected with the ELISA The apoptosis of PBMC was higher in hepatitis B than in control group (P<0.01) . There were significant of the increasing apoptosis in hepatitis B when the cultured time was lengthened (P<0.01) . The level of IL-2, IFN-γ was lower in hepatitis B than that in control group (P<0.01) , with the cultured time lengthening, the level of IL-2, IFN-γ slightly decreased while there were no significant (P>0.05) .The level of IL-4、IL-10 was higher in hepatitis B than that in control group (P<0.01) . And the level became higher with the cultured time lengthening, in addition there was significant (P<0.01) .The cell-mediated immunity was weakened because of the strengthened apoptosis of PBMC and imbalance of Th1/Th2 in chronic hepatitis B, lymphocyte apoptosis and imbalance of Th1/Th2 have somewhat function in the pathogenesis of chronic hepatitis B.
