The Affect on the Biologic Activity of Hepatoma SMMC-7721 cell line of Negative Air Ion In Vitro and related Rignial Mechanism

摘要: 目的探讨空气负离子对体外培养的肝癌SMMC-7721细胞生物学活性的影响。方法将细胞随机分成3组:阴性对照组、空气对照组及负离子治疗组。治疗组根据空气负离子作用时间分成1h组、2h组、4h组和8h组。对所有细胞进行TUNEL染色检测细胞凋亡,流式细胞仪检测细胞的周期变化应用Westernblot方法检测磷酸化P38MAPK的表达变化。结果与阴性对照组和空气对照组比较,空气负离子治疗组细胞凋亡率明显增加,S期细胞所占有的比例明显下降,磷酸化P38MAPK表达明显上升,差异均具有统计学意义(P<0.05)。结论空气负离子通过细胞周期阻滞而使肿瘤细胞发生凋亡,这种效应是通过P38MAPK这条信号通路传导的。Abstract: Objective To investigate the affect on the biologic activity of hepatoma SMMC-7721 cell line of negative air ion in vitro.Methods Dividing the cell into three groups:negative control group, air control group and treatment group.Dividing the treatment group into 1h、2h、4h and 8h group according to action time of negative air ion, all the cells in each group were detected apoptosis and cell cycle with flow cytometry method, the phospho-P38MAPK expression was detected by Western blot assay.Results The apoptosis rate increased, the proportion of S stage cell decreased and the expression of phospho-P38MAPK increase obviously vs negative and air control group, the difference show significance (P<0.05) .Conclusion The negative air ion can induce hepatoma SMMC-7721 cell apoptosis through arresting the cell cycle, such effect was mediated by P38MAPK pathway.
